Matthijs de Ligt's own goal in the 10th minute gave Inter Milan an early lead, but Cristiano Ronaldo's superb free kick in the 68th minute restored parity for Juventus.
With the match heading into penalties, 21-year-old Merih Demiral scored the winning penalty for Juventus, securing an incredible win for coach Maurizio Sarri.
Sarri, who recently joined Juventus from Chelsea, had previously helped the London club win the 2018/19 Europa League title.
Earlier, there were reports of a misunderstanding between Ronaldo and Sarri during Juventus' match with Tottenham in the International Champions Cup.
